Observing regulatory proteins in Ras and Rho signaling by cryo-EM
Ras and Rho families of GTPases are molecular switches which rely on their binding to either GTP or GDP to direct their activity. Interconversion of Rho or Ras GTP to GDP and inversely is a slow process which relies on regulatory proteins, GTPase activating proteins (GAPs) and Guanidine exchange factors (GEFs) to accelerate their activation or inhibition. Using cryo-electron microscopy as our main approach, we shed light into the mechanisms undergirding the regulation of these regulatory proteins. Here, we will present the case of study of two proteins involved in small GTPase regulator regulation: the large tumor-suppressor protein Neurofibromin and the cytoskeletal regulator P-Rex2. We elucidated a new mechanism for the regulation of neurofibromin and were able to characterize the effect of multiple disease-causing mutations. These results exemplify the complex nature of regulatory proteins in GTPase signaling.
